The targeting construct introduced upstream of the lncRNA a loxP site and FRT site upstream of the lncRNA and an additional loxP site toward the 3 prime end of the lncRNA. The endogenous Trp53 response element within the Cdkn1a promoter remains intact. Expression was almost completely abrogated. (J:259337)
