The following elements were inserted into the locus to create an RMCE (recombinase-mediated cassette exchange)-ready allele: a splice acceptor site, an ATG translation start site, an F3 site, a puromycin resistance gene cassette, a flpo recombinase gene cassette (with PGK promoter and poly(A) signal sequence), an FRT site and chicken beta-globin locus insulator sequence. When cells containing this allele are presented with a compatible RMCE vector, the sequence between the F3-FRT sites will be replaced with the sequence between the F3-FRT sites in the vector.
