A loxP site was inserted approximately 400 bp upstream from the start of Mir222 pre-miRNA. A neomycin cassette, second loxP site, and IRES-eGFP were inserted approximately 200 bp downstream from the end of Mir221 pre-miRNA. The neo cassette was excised via flpe-mediated recombination. Cre-mediated recombination deleted Mir221 and Mir222. (J:305774)
