The targeting vector contains a loxP-flanked neomycin inserted into intron 8 and a glutamate to lysine amino acid substitution at position 455 (E455K; GAT to CAT) inserted into exon 8. Cre-mediated recombination removed the neomycin cassette. The mutation corresponds to human tumor-associated missense mutations in the ankyrin domain that disrupts BARD1 recognition of H4K20me0 (histone H4 unmethylated at lysine 20). (J:94077)
