A loxP site, a neomycin resistance gene cassette, and a second loxP site, were inserted 5' to exon 5, and a third loxP site was inserted 3' to exon 5. Exon 5 encodes the end of the second transmembrane domain (TM2) and a portion of the intracellular loop between TM2TM3. Deletion of this exon results in a preterm stop codon and complete KCC2 knockout. The neomycin cassette was removed via Cre-mediated recombination in the germline of mice, leaving loxP sites flanking exon 5. (J:292640)
