The targeting construct replaced exon 2 with a tTA and a floxed neomycin resistance cassette driven by a tetracycline response element and CMV promoter. A third loxP site was inserted between exon 2 and 3. Cre-mediated recombination removed the selection cassette, tetracyline response element, and a portion of exon 2. (J:321349)
