The mouse promoter drives B cell-specific expression of the human MYC coding sequence (exons 2 and 3; Genbank vector MN894515). A codon mutation generates a STOP codon at the third codon (TCG to TAG) and prevents expression of MYC. The codon substitution created a DGYW motif, a known preferential target sequence for somatic hypermutation, that can activate MYC translation following sporadic reversion. Lines 11 and 24 (20 and 8 copies of the transgene, respectively, inserted chr11:6965780-7030487 with translocation of chr 7:68557535-68718226 sequence and chr17:91535994-91546658, mm39, respectively) were generated.
