The targeting construct consisting of a CAG promoter, a loxP-flanked neo stop cassette, a rox-flanked stop cassette, and DREADD (Designer Receptors Exclusively Activated by Designer Drugs) receptor hM3Dq, followed by 2A-NLS-ZsGreen and pA signal all inserted into the Gt(ROSA)26Sor locus was used to generate the mouse line by homologous recombination in ES cells. CAG promoter drives expression of hM3Dq and ZsGreen only after Cre and Dre mediated excision of the floxed and roxed stopper elements. (J:314806)
