The targeting construct is designed to insert a loxP site upstream of exon 4 and a second loxP site, followed by an FRT-flanked neomycin downstream of exon 4. Removal of exon 4 results in a frameshift and the premature truncation of protein. Flp-mediated recombination removed the FRT-flanked neo cassette. (J:314542)
