A fragment of the human alpha-synuclein 1-130 (an N-terminal fragment generated by aberrantly activated asparagine endopeptidase which cleaves alpha-synuclein at N103 and which is prone to aggregate and cause neurotoxicity) cDNA with its 5' UTR containing a Kozak enhance sequence before the initial codon and the 3' polyA tail is under the control of the neuron-specific Thy1 promoter. Western blot analysis confirmed expression of the 1-130 fragment in the brain and showed that the insoluble fragment is enriched in the olfactory bulb, cortex, striatum, substantia nigra, and pons. Insoluble alpha-synuclein phosphorylated at Ser129 is detected within all areas of the central nervous system. Two founders with high expression were used to generate lines.
