Exon 5 was replaced with a modified version of the equivalent, but 11 codons longer, exon from the human gene (exon 6). This exon codes for the binding epitope for the OKT3 anti-human CD3E antibody. The modification to the human sequence is the change of an arginine codon at the 3' end of the human exon to the mouse lysine codon. An FRT site flanked puromycin resistance gene cassette that was inserted into an intron, was removed through subsequent flp-mediated recombination. (J:305166)
