A mutation was engineered in exon 7 to change aspartic acid codon 285 (GAT) to an alanine codon (GCC)(p.D285A) and an FTR site flanked puromycin resistance gene cassette was inserted into intron 6. The puro cassette was removed through subsequent flp-mediated recombination. The mutation abolishes the O-GlcNAc-related enzymatic activity of the encoded peptide. (J:304809)
