CRISPR-targeting inserted the EGFP reporter gene and a 2A peptide as in-frame gene fusion long-single strand ODN (lsODN) structure in exon 1 near the start codon. One pup carrying EGFP in the Ucp1 locus (#11) was obtained and established. DNA sequencing of the offspring showed that insertion resulted in a frameshift mutation of the Ucp1 gene. Western blot of homozygous mice confirmed the absence of Ucp1 protein. (J:310472)
