A loxP site was inserted upstream of exon 1 and a neomycin resistance gene cassette and a second loxP site were inserted into intron 1. Exon 1 was deleted through subsequent cre-mediated recombination. Western blots and RT-PCR confirmed the lack of expression from the knockout allele. (J:310228)
