The following elements were inserted into the ROSA locus: the CAG promoter, a loxP site flanked STOP cassette, sequence for an N-terminal 3xFlag tag in-frame with a codon-optimized mouse Hoxa5 cDNA, poly(A) signal sequences, IRES sequence, the GFP fluorescent marker gene, a WPRE element, poly(A) signal sequences and a neomycin resistance gene cassette. Only after cre-mediated removal of the neo-STOP cassette will this allele express the Hoxa5 and GFP genes. (J:306314)
