An FRT site flanked neomycin resistance gene cassette and a loxP site were inserted upstream of the gene and a second loxP site into intron 1. The neo cassette was removed through subsequent flp-mediated recombination, leaving a conditional-ready allele with exon 1 floxed. (J:307345)
