Exons 2-4 were flanked by loxP sites and an FRT-flanked neomycin-resistance cassette (PGK-neo) was inserted down stream of exon 4 inside the loxP sites. The selection cassette was removed via cre-mediated recombination leaving exons 2-4 floxed. (J:306874)
