A loxP site and an FRT-flanked neomycin resistance cassette were inserted upstream of exon 2 and an F3-flanked puromycin resistance cassette and second loxP site were inserted downstream of exon 2. Flp-mediated recombination removed the selection cassettes, leaving exon 2 floxed. (J:305034)
