Lox2372 and loxP sites were inserted in intron 2, an mCherry and neo cassette flanked by FRT3 and FRT sites was inserted in intron 3, followed by an antisense oriented conditional inversion (COIN) module consisting of a splice acceptor site and a T2A-GFP-pA cassette, and a second set of lox2372 and loxP sites. The two lox sites are in reverse complementation orientation, thus cre-mediated recombination will irreversibly invert the intervening sequence and also delete exon 3. Flp-mediated recombination removed the mCherry and neo selection cassette. (J:302856)
