CRISPR/cas9 genome editing is used to change AAA AAG to GCT GCC, resulting in two lysine to alanine changes at amino acids 63 and 64 (K63A and K64A) in exon 5. Two mismatched bases with silence mutations (CGTG to TGTC) that generate a novel Mwol restriction site were mutated directly upstream of amino acid 64.
