A loxP site flanked STOP cassette was inserted into intron 1 using an sgRNA and a donor plasmid with CRISPR/Cas9 technology. This creates a null allele that can only be transcribed full-length after cre-mediated removal of the cassette. Fluorescent in situ hybridization (FISH) experiments on liver confirmed the absence of transcription from this allele. (J:299830)
