Site-specific PhiC31 integrase-mediated transgenesis is used to insert the targeting construct containing an attB site, the CAG promoter, a loxP flanked PGK-neomycin STOP cassette, a FLAG-tagged mouse Met cDNA, a polyA signal and a second attB site into the intergenic site 2 locus. This construct was microinjected, along with the PhiC31 integrase mRNA into the pronuclei of zygotes of an FVB/N mouse that contains an attP docking site at the H11 locus which is either the Igs2tm3.1Luo or the Igs2tm4Luo. The PhiC31 integrase catalyzes recombination between the attB and attP sites. (J:268934)
