A loxP site was introduced into intron 2 and a loxP and FRT site flanked neomycin resistance gene cassette plus duplicate sequence of the 3' end of exon 10 were inserted downstream of exon 10. The neo cassette was removed through subsequent flp-mediated recombination, leaving a conditional-ready allele with exons 3-10 floxed. (J:298850)
