A 1.2-kbp fragment of genomic DNA containing exons I and II of the protrudin gene was replaced with a PGK-neo-poly(A)-loxP cassette. RT-PCR analysis of mRNA derived from regions corresponding to exon II or exon X demonstrated that the abundance of these transcripts was significantly reduced or nearly undetectable. (J:303874)
