TCRalpha chain variable region (VJ) and beta chain variable region (VDJ) cDNAs are amplified from a secretory protein derived from M. tuberculosis. The ESAT-63-15 peptide-specific CD4+ T cell hybridoma clone 7 expresses TCR chains Vbeta10b-Dbeta1.1-Jbeta2.4 and Valpha11.2-Jalpha27. The complete TCR alpha and beta chains are excised and cloned into VA hCD2 cassette vectors which uses the human CD2 promoter to drive the expression of cDNAs in T cells. (J:141135)
