The diphtheria toxin receptor (DTR) gene, preceded by an artificial intron and followed by bovine growth hormone gene poly(A) signal sequence, and an FRT site flanked neomycin resistance gene cassette were inserted into the endogenous ATG start codon. The neo cassette was removed through subsequent flp-mediated recombination, leaving a null allele that expresses a gene that causes cell death when exposed to diphtheria toxin. (J:243655)
