A loxP site was inserted upstream of the coding region. A loxP and a FRT-flanked neomycin selection cassette were inserted downstream of the coding region. Flp-mediated recombination removed the neomycin selection cassette, leaving the loxP sites flanking the coding region. (J:172134)
