Using a BAC from the RPCI-23 library for the targeting vector, the endogenous genomic sequence, starting from the translation initiation site in exon 1 and ending with exon 2, was replaced with a human PTGES mini-gene consisting of the full-length cDNA with human intron 1 inserted between the exon 1 and 2 cDNA sequences. An F3 site flanked puromycin resistance gene cassette was inserted downstream of the mini-gene. This puro cassette was later removed through flp-mediated recombination. (J:294101)
