A loxP site was inserted into the 5' UTR (12 bp upstream of the ATG start codon) and a second loxP site and an FRT site flanked neomycin resistance gene cassette into intron 3. The neo cassette was removed through subsequent flp-mediated recombination, leaving a conditional-ready allele with exons 1-3 floxed. (J:292694)
