An optimized internal ribosome entry sequence (IRES) fused to an enhanced green fluorescent protein (eGFP) sequence, followed by a FRTflanked neomycin selection cassette, was first inserted 3 bp after the Pomc stop codon within a Pomc gene. Heterozygous mice for the targeting event were crossed with mice expressing Flp-recombinase in the germline to remove neo cassette flanked by FRT sites. (J:291152)
