The targeting vector is designed to insert, from 5' to 3', a loxP site, exons 2-4 of the gene, a STOP cassette, an FRT-flanked neomycin resistance cassette, and a second loxP site downstream of the endogenous exon 1 of the Klf5 gene. Endogenous exon 2 was modified to harbor a mutation at codon 358, resulting in a lysine to arginine mutation (K358R). Flp-mediated recombination removed the FRT-flanked neo cassette. Cre-mediated recombination removed the the inserted exons 2-4 and the STOP cassette allowing the expression of the K358R mutation. (J:286287)
