Exons 4 to 9 were flanked by loxP sites and an FRT-flanked neomycin selection cassette was inserted downstream of the second loxP site. Flp-mediated recombination removed the selection cassette. Cre-mediated recombination deleted exons 4-9 in the germline. Western blot analysis indicates loss of the two highest molecular weight isoforms (C-1, C-2, N-1, and N-2 bands) and a reduction in other isoforms (bands C-3, C-7, and N-3 on the blot) indicating a complex alteration of isoforms.
