The transgene construct consists of an enhanced GFP (EGFP) gene carrying at its 5' an HA epitope followed by a nuclear localization signal and, at its 3' end, stop codons, and a polyA addition site, was used as a reporter. The coding sequence of the EGFP gene was fused, at its initial ATG, to the first ATG of the first exon (and 6.7-kb 5 flanking sequence) of the Kit gene. Construct 3 used in this line also included a 4.5-kb XbaI-BamHI fragment of the first intron. (J:277552)
