A loxP site was inserted upstream of exon 2. An FRT-flanked neomycin resistance cassette, mutant loxP site, inverted 2A-EGFP-WPRE-pA cassette with a splice acceptor, loxP site and mutant loxP site were inserted downstream of exon 2. Flp-mediated recombination removed the selection cassette. Cre-mediated recombination removed exon 2 and inverted the EGFP reporter. (J:287134)
