A loxP site was inserted into an Xmn1 restriction site located approximately 900 bp upstream of the first coding exon and a second loxP site along with a PGK-neomycin resistance cassette flanked by FRT sites was inserted into an AgeI restriction site located approximately 350 bp downstream of the first coding exon. The selection cassette was removed via Flp-mediated recombination leaving exon 1 flanked by loxP sites. The floxed region containg exon 1 was deleted via cre-mediated recombination in the germline.
