A glutamine to X nonsense mutation at amino acid 552 (Q552X) was inserted into exon 7 and a loxP-flanked PKG-neomycin resistance gene cassette was inserted into intron 6. The selection cassette was removed via cre-mediated recombination. Western blot analysis shows that a truncated protein is not detected in mouse embryonic fibroblasts. (J:269011)
