The human NAT2 reference wild-type (NAT2*4) gene is under the control of an approximate 2.3-kb mouse albumin enhancer/promoter. The transgene inserted in intron 3 of the mouse Pde8b gene. A recombination event during insertion caused transposition of most of the albumin enhancer region to the 3 end of the transgene construct, but does not markedly impair expression of the human transcript in the liver. Real-time qPCR indicates that transgenic mice have an approximate 40-55% decrease in Pde8b expression in the brain. Real-time qPCR also indicates that the human transcript is detected in the liver and is 2-fold higher in male liver than in female liver and Western blot analysis confirmed expression, with higher level of protein in the male liver than female liver.
