A Rosa-CAG targeting vector was designed to insert cassette containing (from 5' to 3') a CMV-IE enhancer/chicken beta-actin/rabbit beta-globin hybrid promoter (CAG), rox-site, loxP-site, STOP cassette (with stop codons in all 3 reading frames and a triple polyA signal), second loxP-site, ZsGreen sequence (an enhanced green fluorescent protein engineered for brighter fluorescence and higher expression in mammalian cells), second STOP (adapted from the polyA sequence between mT (membrane-targeted tdTomato) and mG (membrane-targeted eGFP) of pCA-mTmG), second rox-site, tdTomato sequence (a non-oligomerizing DsRed fluorescent protein variant with a 12 residue linker fusing two copies of the protein (tandem dimer)), WPRE (woodchuck hepatitis virus posttranscriptional regulatory element)-polyA, and Frt flanked neomicyne selection cassette (CAG-rox-loxP-Stop-loxP-ZsGreen-Stop-rox-tdTomato-polyA-Frt-Neo-Frt) into the Rosa26 locus.
