Two A to T nucleotide substitutions were introduced resulting in a tyrosine to phenylalanine change at amino acid 448 (Y448F) and at amino acid 537 (Y537F). A FRT-flanked neomycin resistance cassette was inserted upstream of the two mutations. These two mutations abrogate iron binding in the C lobe of transferrin. (J:282105)
