Col1a1^{tm2(tetO-MAPT)Mdk}

The targeting construct includes a Tet-responsive element (TRE or tetO) and mouse prion protein sequences (PrP or Prnp) directing expression of the 4-repeat (0N4R) isoform of non-mutant human microtubule-associated protein tau (MAPT). The construct, a "Flp-In" promoter cassette (PGK-promoter-ATG-FRT) and a pCAGGS-FlpE plasmid were electroporated into modified embryonic stem (ES) cells that had previously been targeted with a Col1a1 "Flp-In" integration cassette (Col1a1-FRT-hygro-pA).
