This allele was generated from a vector that contains two TC>GA mutations (in exon 24 and 26) that create serine to aspartic acid phosphomimetic mutations that lead to a constitutively active enzyme: p.S1712D and p.S1836D. The vector has the potential to generate three different alleles (the two single mutations and one double mutation), depending on the location of the cross-over breakpoints during homologous recombination. The other allele that was created is Dicer1tm1.1Glo. The neomycin reistance gene cassette that was inserted into intron 24, was removed through subsequent cre-mediated recombination. (J:278360)
