The ES cells were targeted in the 3' UTR of the Col1a1 locus with a RMCE construct containing F3-PGK-PuroTK-frt. The targeted Col1a1 locus can later be used as a docking site for the introduction of a plasmid, minimally containing a costitutive promoter, the desired transgene sequence and F3/frt sites.
