A loxP site was inserted into intron 4 and a neomycin resistance gene cassette and a second loxP site into intron 5. The floxed exon 5 and the neo cassette were deleted through subsequent cre-mediated recombination. Exon 5 contains a transmembrane domain and its deletion causes a frameshift, creating a null allele.
