The transgene construct, plasmid PRPGS00048_A_H09 (KOMP project), was electroporated into ES cells. Only clone 2B9 showed proper recombination and was chosen to establish the mouse line. The allele comprises the following elements: an FRT site, a gene trap cassette (mouse En2 splice acceptor, IRES, LacZ gene, SV40 poly(A) signal sequences), a loxP site, a neomycin resistance gene cassette, a second loxP site and a aecond FRT site in intron 2, and a third loxP site in intron 5.
