A ~3 kb genomic fragment, containing part of the Myh6 (myosin, heavy polypeptide 6, cardiac muscle, alpha) regulatory region, was used to drive the cardiac muscle expression of the bacterial chloramphenicol acetyl-transferase reporter gene. The genomic fragment contains sequence from an intergenic SphI restriction site to the MaeIII site at 1070..1074 in exon 3 and was generated by ligating a fragment from the SphI site to an NdeI (at -42..-37 relative to the transcription start site) fragment into the NdeI site at -42..-37 close to the 5' end of Tg(Myh6-cat)alpha-1.3Rbns The sequence contains the following elements: ~1.5 kb of upstream sequence (containing CCAAT and TAATA boxes and other promoter sequences), and the 5' end of the Myh6 gene (comprising the first two (5' UTR) exons and introns, and the non-coding part of exon 3). Chloramphenicol acetylation activity was only detected in the heart and not in aorta, breast, biceps, diaphragm, kidney, liver, brain, uterus, thigh, calf or spleen.
