The transgene comprises the following elements: the mouse Myh6 promoter, sequence for the human Myc epitope tag immediately after the ATG start codon, the wild-type mouse Mybpc3 cDNA, and human GH gene poly(A) signal sequence. Three mouse lines (3/1, 1/3, 2/1) were established; line 2/1 was the highest expressing line (approximately 4.5x endogenous mRNA level) and was used for further experiments. (J:50237)
