The transgene construct comprises the mouse myosin, heavy polypeptide 6, cardiac muscle, alpha promoter, modified yellow fluorescent protein gene sequence, and SV40 poly(A) signal sequences. The EYFP gene was modified as follows: 5' yeast COX4 sequence (coding for LSLRQSIRFFK) was added to the 5' end to direct the sequence to mitochondria; the original 5' end of the EYFP gene was placed downstream of the 3' end connected through a linker sequence (coding for VDGGSGGTG). In this transgene the resulting mt-cpEYFP acts as a superoxide biosensor in cardiac mitochondria. (J:274665)
