A modified mouse Nrg1 type IIIb1a cDNA, plus sequence for two hemagglutinin (HA) peptide tags inserted immediately after the ATG start codon, was placed in a Thy1 expression vector. This vector contains the entire Thy1 gene (including 5' promoter and 3' polyA signal sequences and neuronal-specific expression signal containing introns) with the coding region removed (from the ATG in exon 2 to the end in exon 4, including intervening introns 2 and 3). Nrg1 sequence was inserted between the remaining sequences of Thy1 exons 2 and 4. The Nrg1 modification entailed the insertion of a stop codon after the Gly-Ile-Glu-Phe (GIEF) codons 289-292 and truncation of the cDNA a short distance after. The transgene was injected into fertilized C57BL/6 oocytes. The premature stop codon results in a peptide that mimics the product that results from proteolytic cleavage of Nrg1 by Bace1. (J:262505)
