The targeting vector was designed to insert a loxP site upstream of the entire coding region. A second loxP site and a frt site, followed by neomycin selection cassette (neo), a third loxP site, and a green fluorescent protein (GFP) sequence followed by a second frt site, were inserted downstream of the gene.
