A loxP site was inserted upstream of exon 1, and an frt flanked neomycin cassette and another loxP site was inserted downstream of exon 1 by homologous recombination. The neomycin cassette was removed through Flp-mediated recombination resulting in an allele with a floxable exon 1. Western blot on extracts derived from brain of homozygous animals, following Cre-mediated excision of exon 1, did not detect any protein product. (J:235517)
